There was the very real possibility that Killua might not just become friends with Gon, but they could one day fall in love. That was the threat Gon represented to Illumi. Was it a general comment meant to describe the two of them playing a cat and mouse game?
That would make sense given they have a standing contract to kill each other. Illumi has good reason to want Gon dead. Best to let someone else handle it. There are several places in the story where we saw Illumi take the initiative to kill Gon, but was stopped for one reason or another.
One of those reasons being Hisoka.
Illumi is hopeful that Hisoka will eliminate the Gon threat. But several times Hisoka seems to drop the ball. Doubtless, Illumi was disappointed by that. The next time was during the Yorknew arc when Gon and Killua were captured by the Troupe. Hisoka had another opportunity to kill Gon, but seemed to be more a threat to Killua.
The next time was on Greed Island, once again Gon is presented to Hisoka on a silver platter. Hisoka had ample opportunity to kill Gon. And yet he did not. How frustrated Illumi had to be. Along with the bonus of eliminating the threat of Alluka. For this Illumi uses Hisoka, but once again things do not go according to plan. Hisoka sorta handles the butlers, but not much else. Illumi is the clear looser here, with Hisoka pleased as punch that Gon will survive to fight him one day in the future as was always expected.
Illumi wants Gon dead, as soon as possible. Hisoka wants to kill Gon, but not for a long time yet.
Gon is thus caught in the middle along with Killua. When did Illumi and Hisoka figure out Killua was in love with Gon? Although Illumi feared the possibility that Killua might become more-than-friends with Gon right from the very beginning, a series of events worked to reinforce that notion. Eventually there came a moment when Illumi was convinced Killua was in love.
Want to guess when that was?
In the fateful scene Anime Ep. Killua discovers the needle implanted in his mind long ago by Illumi. He removes the needle and instantly regains full control of himself and his abilities. Rammot continues to taunt him to which Killua reflects on his relationship with Gon, how Gon is his most precious friend, and proceeds to destroy Rammot.
Consider the situation had Rammot not mentioned Gon. Or had it been any other Ant who had no knowledge of Gon but was still as powerful as Rammot. In those conditions Killua would have submitted to his brainwashing and fled the more powerful opponent in keeping with his training. A sound tactic since it would have lead the Ant away from the area, reducing the chances of it finding Gon. That needle in his mind served as a handicap, it was what prevented him from defeating Shoot.
But the turning point was Gon. Had Shoot been a direct threat to Gon, doubtless Killua could have summoned the strength to discover the needle and defeat Shoot.
It all comes down to circumstance and love. But that needle is critical to Illumi. It gives him great control over Killua, reinforcing all the assassin training and most of all the special restriction against Killua making friends. That needle provides Illumi with information about where Killua is, what his mental and emotional state are, and may go as far as transmitting images.
Specifically, that Killua was impowered by his love for Gon. Even if this was not the moment, a while later, following the battle of the Chimera Ants, the Zoldycks would have been briefed on the outcome. They would have known that Gon was in a coma and not expected to live. Soon after, Killua vows to save Gon and returns to the Zoldyck mansion. That was all the proof the family needed to know Killua was in love, and double so for Illumi.
Milluki even remarks that Killua is different than them, that he cares for people.

The only way for Illumi to save himself was to see Killua dead or Gon. Having utterly failed to kill Alluka or Killua or even Gon, and after being teleported back home by Something at Killua's command, poor Illumi probably had some tuff questions to answer by Silva.
Pity we missed that chat. However Illumi's current standing in the family, with Silva in particular is unknown. Assuming things are not great for Ilumi it may suggest why he took Hisoka's invatation to join the Spiders. The Troupe could provide him with funds but the big bonus was they were going on a voyage away from Silva's reach.
But when did Hisoka know Killua was in love with Gon?
When he was younger, Illumi's hair only reached his shoulders. He wore long sleeves with a shirt underneath, as well as grey pants. Illumi, for the most part, displays very little expression. In contrast to his expressionless demeanor, Illumi often speaks in a somewhat joyful and absent-minded tone.
He is generally a stony, ruthless person with an extremely cold way of thinking, reflects only his interest. Still, he holds a twisted and overprotective form of love for his younger brother, Killua; this manifests to the point that he used his Nen ability to control Killua and ensure he survives and obeys.
He and his father raised Killua to believe that the only thing sustaining him was darkness, and that he could only obtain joy in causing the death of others. To ensure that Killua would live to carry on the family business, and as a means of protecting his little brother, Illumi implanted a needle in Killua's brain, forcing the latter to run from battles he wasn't sure he could win. Illumi once came to the aid of Hisoka by standing for him as a double in the Phantom Troupe's hideout while Hisoka attempted to find Chrollo.
However, when Hisoka asked Illumi would it be okay if he killed Killua, Illumi displayed a horrifying side of himself and asked if Hisoka wanted to die then and there - he acted very similar to how Hisoka did himself after Illumi was opting whether or not to kill Gon, whom he sees as a threat due to his closeness with Killua, during the Hunter Exam, albeit on a much lesser expense.
Illumi cares for most of the Zoldyck family members. He is willing to handle Alluka's power, even if it costs him his life. He has stated, however, that family members are not allowed to kill during an inner mission a mission where there is internal conflictbut insinuates that he would be fine with killing Alluka and does not consider Alluka a member of the family.
Tsubone implies that Illumi's sinister ambition to control Alluka's power even caused him to stray from the path of an assassin. He is the best friend of Gon Freecss. Killua has spiky silver hair, very light skin and blue eyes. His eyes change shape depending on the mood that he's in, narrowing and sharpening when he goes into assassin mode.
Killua is fairly lean at the start of the series, due to constant physical conditioning and torture training he received when he was young. As time passes, he becomes more muscular and toned. Tsubone implies that Illumi's sinister ambition to control Alluka's power even caused him to stray from the path of an assassin.
He is always in search for strong opponents, and would spare those who have great potential, such as Gon and Killua. Hisoka's appearance is similar to that of a magician or jester. He is tall and has light skin and a fairly toned stature.
His attire is usually adorned with various suit symbols on the front and back torso, and he changes outfits in each story arc. He wears face paint: In the 13th Hunter Chairman Election arc, Hisoka is shown wearing two earrings with ornamental hearts.
Illumi x Killua
In his first appearance in Nippon Animation's anime series, Hisoka's hair is blue. After a revamp of the show it was replaced with a red, almost neon pink color to be in accordance with the manga. In Madhouse's adaptation, Hisoka has red hair and light amber eyes.
During the Yorknew City arc in the manga, Hisoka dyes his hair light green. These dynamic variations in appearance illustrate the complexity of his character. Hisoka is portrayed as entirely self-serving, self-absorbed, and selfish; he does whatever he likes as long as it pleases him in some way. His insatiable love for spilling the blood of powerful fighters in combat and his enjoyment of extreme pain while doing so fuels his seemingly sadomasochistic desires.
In order for them to grow to their full potential so as to improve the pleasure of killing them laterHisoka allows the main protagonists to live in situations wherein he is easily able to kill them.
